چکیده

The luminescent Eu complex, DTBTA-Eu, is highly stable in terms of ligand-metal dissociation, and can be labelled to amino groups of proteins and other bio-molecules. Owing to the long lifetime of Eu luminescence, time-resolved measurement of DTBTA-Eu removes background fluorescence and enhances the sensitivity of the labelled materials. This property was applied to time-resolved luminescence microscopy and DNA microarray. In these systems the high sensitivity and reliable quantification of the image was proved. In time-resolved images, autofluorescence of tissues and cells was dramatically reduced. In addition, due to the high stability of the metal complex towards photo-bleaching and ozone oxidation, which had been serious problems in the conventional normal fluorescence microscopy and microarray using organic dye labels, deterioration of the images on time elapse turned out to be drastically reduced in the timereesolved imaging, compared to the normal fluorescence images.

Preliminary examination of time-resolved fluorometry for protein array applications.

The advantages of time-resolved fluorometry over conventional fluorometric analysis are well known. However, time-resolved fluorescence has not as yet found wide applications in protein microarray or other multiparametric methods of analysis. Here we describe a general method which is suitable for multiparametric and microarray analysis, based on time-resolved fluorometry. A polystyrene surface...

Design of Novel, Water Soluble and Highly Luminescent Europium Labels with Potential to Enhance Immunoassay Sensitivities.

To meet the continual demands of more-sensitive immunoassays, the synthesis of novel luminescent Eu(III) chelate labels having similar substituted 4-(phenylethynyl)pyridine chromophores in three different chelate structure classes are reported. Significantly enhanced luminescence intensities were obtained, evidently caused by the intra-ligand charge transfer (ILCT) mediated sensitization, but t...

Amine-reactive forms of a luminescent diethylenetriaminepentaacetic acid chelate of terbium and europium: attachment to DNA and energy transfer measurements.

An isothiocyanate form of a lanthanide chelate which is highly luminescent when bound to terbium or europium has been synthesized. The chelate consists of diethylenetriaminepentaacetic acid (DTPA) covalently joined to a chromophore, 7-amino-4-methyl-2(1H)-quinolinone (cs124), and to L-p-aminophenylalanine, in which the aromatic amine was further converted to an isothiocyanate group. Ethylenedia...

Europium and samarium as labels in time-resolved immunofluorometric assay of follitropin.

This time-resolved immunofluorometric assay for human follitropin involves use of europium- or samarium-labeled monoclonal antibodies, with an average incorporation ratio of 3 mol of Eu3+ or Sm3+ per mole of antibody. These lanthanide ions are bound to the antibody molecules by means of the anhydride of diethylenetriaminepentaacetic acid. The solid-phase antibody is immobilized inside polystyre...

Calibration beads containing luminescent lanthanide ion complexes.

The reliability of lanthanide luminescence measurements, by both flow cytometry and digital microscopy, would be enhanced by the availability of narrowband emitting, UV excited lanthanide calibration beads. 0.5-, 3-, and 5-microm beads containing a luminescent europium-complex are manufactured. The luminescence distribution of the 5-microm beads is measured with a time-delayed light-scatter-gat...
